Cooperative activation of the cardiac myofilament: the pivotal role of tropomyosin.

چکیده
The contraction and relaxation of the beating heart are mediated by highly coordinated processes that allow rapid activation and deactivation of the myofilaments. The sarcomere is the fundamental contractile element, and its shortening is the result of the myosin-containing thick filament sliding past the actin-containing thin filament. Fueled by ATP hydrolysis, myosin is the molecular motor that drives sarcomere shortening through a cyclic, ratchet-like interaction with actin. A critical component of cardiac contraction is the regulation of myosin-actin interactions, which are governed predominantly by the thin-filament regulatory proteins troponin and tropomyosin.

Troponin and tropomyosin: proteins that switch on and tune in the activity of cardiac myofilaments.
We present a current perception of the regulation of activation of cardiac myofilaments with emphasis on troponin (Tn) and tropomyosin (Tm). Activation involves both a Ca2+-regulated molecular switch and a potentiated state, dependent on feedback effects of force-generating crossbridges. Recent developments in the elucidation of the structure and arrangement of the myofilament proteins offer in...
متن کاملTropomyosin Flexural Rigidity and Single Ca2+ Regulatory Unit Dynamics: Implications for Cooperative Regulation of Cardiac Muscle Contraction and Cardiomyocyte Hypertrophy
Striated muscle contraction is regulated by dynamic and cooperative interactions among Ca(2+), troponin, and tropomyosin on the thin filament. While Ca(2+) regulation has been extensively studied, little is known about the dynamics of individual regulatory units and structural changes of individual tropomyosin molecules in relation to their mechanical properties, and how these factors are alter...
متن کاملStructural determinants of cooperativity in acto-myosin interactions.
Regulation of muscle contraction is a very cooperative process. The presence of tropomyosin on the thin filament is both necessary and sufficient for cooperativity to occur. Data recently obtained with various tropomyosin isoforms and mutants help us to understand better the structural requirements in the thin filament for cooperative protein interactions. Forming an end-to-end overlap between ...
متن کاملSimulation Analysis of Cardiac Muscle Isotonic Contractions at Different Pre- and Afterloads
Since regulation of cardiac muscle contraction is complex, many simulation studies have been conducted to systematically analyze regulatory mechanisms underlying the force-velocity relationship. However, past studies were performed with models lacking detailed thin filament activation despite its essential regulatory role. Here a novel cardiac muscle contraction model is presented that consider...
متن کاملPredicting Effects of Tropomyosin Mutations on Cardiac Muscle Contraction through Myofilament Modeling
Point mutations to the human gene TPM1 have been implicated in the development of both hypertrophic and dilated cardiomyopathies. Such observations have led to studies investigating the link between single residue changes and the biophysical behavior of the tropomyosin molecule. However, the degree to which these molecular perturbations explain the performance of intact sarcomeres containing mu...
